Multi-ancestry genome-wide association study of 21,000 cases and 95,000 controls identifies new risk loci for atopic dermatitis

چکیده
Genetic association studies have identified 2 loci associated with atopic dermatitis risk predominantly in populations of European ancestry. To identify further susceptibility loci for this common, complex skin disease, we performed a meta-analysis of > 5 million genetic variants in 2 ,399 cases and 95,464 controls from populations of European, African, Japanese and Latino ancestry, followed by replication in 32,059 cases and 228,628 controls from 8 studies. We identified ten new risk loci, bringing the total number of known atopic dermatitis risk loci to 3 (with new secondary signals at four of these loci). Notably, the new loci include candidate genes with roles in the regulation of innate host defenses and T cell function, underscoring the important contribution of (auto)immune mechanisms to atopic dermatitis pathogenesis.

Genome-wide association analysis of more than 120,000 individuals identifies 15 new susceptibility loci for breast cancer
Genome-wide association studies (GWAS) and large-scale replication studies have identified common variants in 79 loci associated with breast cancer, explaining ~ 4% of the familial risk of the disease. To identify new susceptibility loci, we performed a meta-analysis of GWAS, comprising 5,748 breast cancer cases and 8,084 controls together with 46,785 cases and 42,892 controls from 4 studies ge...
متن کاملA genome-wide association study of atopic dermatitis identifies loci with overlapping effects on asthma and psoriasis
Atopic dermatitis (AD) is the most common dermatological disease of childhood. Many children with AD have asthma and AD shares regions of genetic linkage with psoriasis, another chronic inflammatory skin disease. We present here a genome-wide association study (GWAS) of childhood-onset AD in 1563 European cases with known asthma status and 4054 European controls. Using Illumina genotyping follo...

The genetic basis of sporadic colorectal cancer (CRC) is not well explained by known risk polymorphisms. Here we perform a meta-analysis of two genome-wide association studies in 2,627 cases and 3,797 controls of Japanese ancestry and 1,894 cases and 4,703 controls of African ancestry, to identify genetic variants that contribute to CRC susceptibility. We replicate genome-wide statistically sig...
متن کاملAssociation analyses identify two susceptibility loci 5q31 and 5q22.1 for atopic dermatitis in Chinese Han population.
BACKGROUND Atopic dermatitis (AD) and other atopic diseases often share some common genetic and pathogenic bases. Recent genome-wide association studies (GWAS) have identified several loci associated with atopic diseases, allergic sensitization and asthma in different populations. The aim of this study was to investigate whether these susceptibility loci were related to AD in Chinese Han popula...
متن کاملGenome-wide trans-ancestry meta-analysis provides insight into the genetic architecture of type 2 diabetes susceptibility
The majority of GWAS of T2D susceptibility have been undertaken in populations of European ancestry1–5, predominantly because of existing infrastructure, sample availability and relatively poor coverage by many of the earliest genome-wide genotyping arrays of common genetic variation in other major ethnic groups6. However, populations of European ancestry constitute only a subset of human genet...
